CAR T cell therapy is an immunotherapy that can change the patient’s own T cells, allowing them to recognize and attack cancer. T cells are taken from the patient’s blood. Genes for special receptors that bind to a protein on a patient’s cancer cells are then added in the laboratory.

Identification of the target occurs before and is a prerequisite for CAR-T cell therapy. In order to avoid damage to healthy tissues caused by CAR-T cell therapy, the target must be limited to tumor cells. To date, ideal therapeutic effects have been achieved against a variety of tumor-associated antigens (TAAs).

However, it is difficult to find an ideal biomarker as an anti-tumor target, which is highly expressed on the surface of tumor cells, but rarely expressed on healthy cells. More commonly, selected biomarkers are expressed on both tumor cells and healthy cells, with significant expression level discrimination. This is why close attention should be paid to on-target-off-tumor cytotoxicity of CAR-T cell therapy.
